關於羅馬教會的起源，聖經上隻字未提。據經學家的推測，很可能是由曾在耶路撒冷「僑居的羅馬人」（宗2:10），即那些在聖神降臨節日，在耶京聽宗徒之長伯多祿的宣講而歸化的人，將福音的種子帶到了羅馬。可是，伯多祿宗徒在喀勞狄在位時，即公元44年後（宗12:17），或至少在尼祿皇帝登極（公元54年）後，來到了羅馬，立定了宗座，卻是一定的事實。
當保祿寫《羅馬書》時，羅馬教會已日漸興旺，而名聞於世（1:8）。所有的信友，大多數是由教外歸化的（1:13, 11:13, 15:15-16），彼此同心合意，平安相處（15:14, 16:17）。
保祿於公元58年初，在格林多寫了本書（15:25-28）。他早有意要「看看羅馬」（宗19:21），只因東方的傳教工作，一時未能如願以償。當東方傳教工作告一段落時，他遂決意要去西方擴展基督的神國。他想「當我往西班牙去的時候，我希望中途見到你們」，看看羅馬（15:24），所以保祿寫本書信的目的，是為通知羅馬信友，他快要來到羅馬。
本來保祿傳教的原則是：不往別人宣講過福音的地方去宣講福音（15:20,21），但他卻願羅馬信友從他這身為外邦人的宗徒身上，獲得有益神靈的「一些效果」（1:11-15）；為此，他願藉這封書信，先給他們報告一下自己「福音」的綱要：即整個人類，不分猶太人與外邦人，都同樣需要耶穌的救贖。
在保祿所寫的書信中：本書的筆調自成一格，可說是一部特別涉及神學問題的書，道理深奧高超，語調沈重有力，超過其他書信。宗徒所徵引的證據，如此鞭辟入裏，致使讀者無一不受感動。我們只看他惋惜選民大部份被棄的事（9-11章），便可舉一反三了。
本書除引言和結論外，可以分為兩編。作者的命題是：「福音正是天主的德能，為使一切有信仰的人獲得救恩，先使猶太人，後使希臘人」（1:16-17）。首編為教義部份（1:18-11:36）：作者志在說明外邦人和猶太人都離棄了正義之道；眾人之所以能成義，再與天主和好，全是藉着信德；人只能賴信德而成為天主的兒女；可惜身為選民的猶太人，大多數不知或不願走這唯一得救的道路。次編為倫理部份（12:1-15:13）：作者首先說明信友對天主對人和對自己的義務；以後勸他們善用基督徒的自由；最後問候羅馬的友好，並且以讚頌天主的絕妙言辭，結束了他這不朽之作。",
